STUDY OF CARTILAGE AND BONE DEVELOPMENT OF BONYLIP BARB LARVAE (OSTEOCHILUS VITTATUS VALENCIENNES, 1842) BY USING DIFFERENTIAL STAINING ALIZARIN RED S AND ALCIAN BLUE

Bonylip barb (Osteochilus vittatus Valenciennes, 1842) is one of the cultivated freshwater fish which has potential to be developed into superior products in fisheries. Osteochilus vittatus Valenciennes, 1842 is a member of Cyprinid family, alltogether with carp (Cyprinus carpio) and Zebrafish (Dani...

Osteochilus vittatus Valenciennes, 1842 is a member of Cyprinid family, alltogether with carp (Cyprinus carpio) and Zebrafish (Danio rerio). The study of cartilage and bone development in bonylip barb has never been carried out, while in carp and zebrafish have been reported. Therefore, it is necessary to determine the temporal development of the embryo, cartilage and bone in the bonylip barb larvae. This information is important to identify any kind of anomalies that may occur in the fish. The development of the embryos was studied by using inseminated eggs that fertilized by using sperm suspension. The development of the embryos was studied visually by using microscope and photographed. The cartilage and bone development studies were performed by using differential staining of alizarin red S and alcian blue. Fish larva at the ages of 0, 1, 3, 5, 10, 20, 30, and 40 days post-hatching (dph) were obtained from in vitro inseminating the eggs (from stripping) and fertilized those with sperm suspension. Larvae were fixed in 10% formalin solution, then stained with alcian blue and alizarin red S following the method used by Konishi (1999). Developmental observations were carried out using a stereo microscope including cartilage and bone development in (i) the head (cranium), and (ii) the spine (truncus) and fins. The result showed that the embryonic development starts with the division stage that occurs 30 minutes, morula stage occurs 90 minutes, blastula stage occurs 2 hours, gastrula stage occurs 4 hours and organogenesis occurs 6 hours after fertilization. Hatching and depletion of the yolk occurred 24 hours and 72 hours after fertilization, respectively. The initial cartilage development of bonylip barb larvae takes place at 1 dph and has been fully formed at 30 dph, while the initial development of bones takes place at 20 dph and the vertebrae are fully formed at the age of 40 dph. text
