One example of skin care products that are widely used is day cream. The increasing need for day cream must be accompanied by an increase in product quality and safety. Cosmetic products are very susceptible to contamination by microbes, which can reduce the quality and efficacy of these products. Therefore, to reduce the potential for product damage due to microbial contamination, safe and effective preservation methods are needed, one of which is the addition of antimicrobial preservatives to inhibit microbial growth in day cream products according to national standards. In this study, the efficacy test was carried out for 3 variations of the formula sample day creamX, each contain different preservative concentrations through a microbial challenge test based on the European Pharmacopoeia, Indonesian Pharmacopoeia, and BPOM references which are generally used by the national cosmetic industry. The preservative used is a combination of phenoxyethanol and chlorphenesin. Day cream formulas X variations 1, 2, and 3 contain high, medium, and low preservative concentrations, respectively. This study aims to determine the status of compliance with European Pharmacopoeia, Indonesian Pharmacopoeia, and BPOM standards for the day cream formulas X with various antimicrobial preservative concentrations, and to determine the preservative concentration formula sample day creamX which has the best efficacy. The research method was started by making a growth curve for each challenge test microbes, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Candida albicans, and Aspergillus niger. Then the abundance of microbial contaminants of the three formula day cream X were calculated with Total Plate Count and microscopic observation of bacteria with Gram stain, then a single culture challenge test was carried out for each challenge test microbes. The results showed that the optimal inoculum age of E. coli; P. aeruginosa; S. aureus; C. albicans; and A. niger are 4 hours respectively; 8 hours; 8 hours; 8 hours; and 96 hours. The three sample formulas for day cream X intact meet the limits of contaminant microbial contaminants according to the BPOM and European Pharmacopoeia references (<3×103 CFU/mL) with the number of contaminant bacteria in each sample formula of day cream Xvariation 1; 2; and 3 are 1.3×103 CFU/mL, respectively; 1.78×103 CFU/mL; and 2.53×103 CFU/mL where the contaminant microbes were Gram-positive bacteria in the form of bacilli. In addition, based on the results of the microbial challenge test, the sample formula day cream X variation 1 gave a log reduction in the number of microbial cells was higher than variation 2 and 3, and the three sample formulas for the day cream X passed the acceptance standards of European Pharmacopoeia, Indonesian Pharmacopoeia, and BPOM. It can be concluded that the formula concentration of preservative in the sample of day cream X has the best efficacy among the three formula samples is the formula sample day cream X variation 1 which contains a high concentration of preservative. text
