Summary:Kabau fruit peel has higher antioxidant activity than kabau fruit. Oxidative stress may cause immunodeficiency. The administration of antioxidant can changes immunological and oxidative stress parameters, antioxidant can enhance the imun system. The purpose of this study was to determine the immunomodulatory activity of 96 % ethanol extract and 50 % ethanol extract from the peel of kabau fruit and to isolate the secondary metabolites from the peel of the kabau fruit. Total phenol and flavonoid contents were determined by a UV-Vis spectrophotometer. Immunomodulatory activity was tested using the carbon clearance method. Eighteen male Swiss Webster mice were divided into groups, given test substance (96 % ethanol and 50 % ethanol extracts) at dose 100 and 200 mg/kg, 0.5 % Na- CMC as a negative control and 19.5 mg/kg BW Stimuno as a positive control. The treatment was given for seven days. The active extract was further fractionated and isolated using chromatography. Indentification and characterisation of the isolat using TLC-Scanner and NMR. The total phenol and flavonoid contentss of ethanol 96 % extract were 9,507 % and 1,850 %, the total phenol and flavonoid levels of ethanol 50 % extract were 10,512 % and 0,593 %. The results show that 96 % ethanol extract of kabau fruit peel had immunostimulant activity, with a phagocytosis indeks of 1.17. Isolation of the 96 % ethanol extract of kabau fruit peel resulted two isolates, quercitrin with Rf value of 0.55 and a terpenoid compound (Ab-02) with Rf value of 0.66.
