Summary:Preservatives most used nowadays often induce allergic reactions and another hazardous effect in human. Propylparaben is one of the most used preservatives in cosmetics. Unfotunatelly, there has been claim that paraben can induce cancer cell growth which relates to estrogen. Thus alternatives for preservatives in cosmetic use alone is be explored. One of the solution is using extract from plants that known to have potent antimicrobial activity. Pomegranate fuit (Punica granatum L. ) has been used as antiparasite and antiworms and researched have antimicrobial activity. This research was carried out to determine antimicrobial activity and the effectiveness as preservative from three cultivars of pomegranate fruit peel which were the red, black, and white, prepared as ethanol extract, ethyl acetat fraction, hexane fraction, and aqueous fraction. Preliminary test of antimicrobial activity used agar diffusion testing to extract, and the MIC is determination with microdilution method in 96-microwell plate, and followed by MBC determination. Sampel for simulation to test the effectivness preservative is cream with oil in water phase. Effectiveness of preservative is conducted by counting the number of colony in serial dilution at physiological NaCl. The result showed that there was no significant differences in activity among three cultivars, but the black had the higest activity. The highest MIC was found in ethanolic extract of black and red pomegranate peels against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The most active fraction was ethyl acetate fraction of black and red pomegranate peels against Staphylococcus aureus. Effectiveness of preservative testing was conducted with ethanolic extract of black pomegranate at 0,5%;1%;2%. Ethanol extract of black pomegranate peel at 2% could be an alternative preservative in cosmetics.
