xanthine and then to uric acid. Excessive deposits of monosodium urate in joints and bones will
result in an acute arthritis attack called a gout. Traditionally, the galangal rhizome (Alpinia galanga
(L.) Willd.) is used as a joint pain medication. Therefore, scientific testing is needed to verify the
efficacy of the plant as a joint pain medication through inhibition of xanthine oxidase activity. This
test was performed on ethanol extract, essential oil-free water extract, and the volatile oil of fresh
rhizome Alpinia galanga (L.) Willd. Inhibition of xanthine oxidase activity was determined by
measuring the levels of uric acid formed by the reaction between the substrate (xanthine) and the
enzyme (xanthine oxidase), compared with the reaction beetwen the substrate (xanthine) and the
enzyme (xanthine oxidase) when the sample was added by using a UV-Vis spectrophotometer at a
wavelength of 284 nm. The ethanol extract had the greatest inhibitory activity at 100 µg/mL
concentration of 16.28 ± 1,23 (%). The IC??value of ethanol extract was 669.70 µg/mL, while
allopurinol had IC??of 1.54 µg/mL. Based on the results of this study, it showed that fresh galangal
rhizome (Alpinia galanga (L.) Willd.) that are used traditionally has inhibitory activity of xanthine
oxidase, so it can be said there was alignment between test result with traditional use as a joint pain
medication.
