Antibacterial drugs have been developed in Indonesia, especially from natural products, include Rosmarinus officinalis, Lagerstroemia speciosa and Dimocarpus longan. The aims of this study was to determine the antibacterial activity of two combination of rusmarin, bungur and lengkeng leaves by in vitro test using microdilution method and in situ test in topical preparations. The crude drug of rusmarin, bungur and lengkeng were extracted using 96% ethanol. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of extract were determined by microdilution method against S. aureus and S. epidermidis. In situ testing, the bacteria were injected into the back skin of rabbit with wounded initiation and then intracutaneous injection and treated by topical preparation until healed after induction. The result showed that the MIC ethanolic extract of rusmarin and bungur leaves were 128 and 512 µg/mL, and MBC were 256 and 1024 µg/mL respectively against S. aureus. The MIC Extract of lengkeng and bungur were 256 and 1024 µg/mL respectively and MBC were >4096 µg/mL against S. epidermidis. Combination of rusmarin-bungur showed synergistic effect against S. aureus and lengkeng-bungur showed additive effect against S. epidermidis therefore combination of rusmarin-bungur extract was continued to be tested in situ on the back skin of rabbit. The ointment healed infection on day 10 significantly while the cream healed infection on day 5 and day 19 significantly compared to the control. The healing time of ointment was 13.33 ± 3.06 days and cream 14.33 ± 5.13 days, while the control showed healing time after 15 days. text
