Summary:Ibuprofen is a Non-Steroidal Anti-Inflammatory Drugs that practically insoluble in water but has high permeability. Therefore performance of ibuprofen in a tablet is determined by the rate of dissolution. This research aims to improve disolution rate of ibuprofen using solid dispersion method in chitosan with several molecul weight variations. Solid dispersion was made by solvent evaporation method at various rasio of ibuprofen-chitosan. Electrostatic binding of ibuprofen-chitosan were optimized including volume ratio of chitosan solution in acid and ibuprofen solution in ethanol. Physical mixture of ibuprofen-high molecul weight (HM) and ibuprofen-low molecul weight (LM) used as comparision to formulated solid dispersion. Solubility dan disolution profile of obtained solid dispersion in HCL 0,1 N pH 1,2, water, and fosfat buffer pH 7,2 was evaluated. Ibuprofen-HM with rasio 1:1,5 showed the highest solubility dan disolution rate in HCL 0,1 N pH 1,2. (p<0,05), in contrast ibuprofen-LM with rasio 1:1,5 showed the highest solubility dan disolution rate in water dan fosfat buffer pH 7,2 (p<0,05). FT- IR spectrum and SEM photograph indicated interaction between ibuprofen and chitosan within obtained solid dispersion.
