Summary:In pharmaceutical industries, physicochemical properties of an API have a lot of impact on the formulation of a drug. Not all API have favorable physicochemical properties to be formulated as a drug. To solve this problem, many method were used such as changing the form of the API to its salt or cocrystal form. In this literature review, the influence of dicarboxylic acid that are used as coformer in cocrystal manufacturing were observed and summarized. Research regarding cocrystal manufacturing has been conducted in the past decade to alter the physicochemical properties of a substance such as solubility, melting point, hygroscopity, permeability, and compressibility. The change in physicochemical properties could ease the manufacturing of a drug and also further increase the stability of an API. Dircaboxylic acid as a coformer will improve the solubility of a cocrystal with the solubility and chemical structure of coformer played a role in it. The hygroscopicity of a cocrystal tend to decrease with glutaric acid and malonic acid is the worst in improving the stability of a cocrystal. Cocrystal could improve the permeability of an API by using the more lipophilic dicarboxylic acid. The tabletability of a cocrystal will improve with the modification of the structure of API that made it possible to plastically deformed.
