Being widely used in fields ranging from medicine, fragrance, flavour, to its potential to become biofuel, terpenoids are in high demand. Efforts have been made to make sufficient supply of terpenoid, one of them is metabolic engineering on bacteria which still requires optimization. This study aims to review the heterologous expression of mevalonate pathway including the challenges and strategy to be applied in order to improve the terpenoid production level. The method used in this research was traditional literature review of published articles obtained from Google, Google Scholar, and PubMed. The selected articles were under research journals and literature review category, written in English, and have Digital Object Identifier (DOI). To ensure that the data obtained are reliable, only those that have Q1, Q2, and Q3 rank based on Scimago are selected. The biochemistry profile was also checked. From 47 reviewed journals, it was discovered that several bacteria have been used as hosts for heterologous mevalonate pathway expression. Other findings include the implementation challenges such as feedback inhibition as well as the influence on other metabolic pathways. The strategy that can be applied to combat these challenges is metabolic flux balancing, such as enzyme overexpression in central pathway, gene selection, and regulation of promoters. As a suggestion, flux balancing for optimization in heterologous metabolic engineering of mevalonate pathway could be further investigated on a laboratory scale. text
