Acute toxicity study of Phyllanthus niruri on hematological test, kidney and liver indicate there is no toxicity, while sub cronic toxicity on testis show there are toxicity that cause hormonal imbalance and degenerative damage. Sertoli cell is one of cell that give huge role on spermatogenesis. Disruption on sertoli cels cause disruption on spermatogenesis that can impact on fertility. Non classical testosterone signalling pathway is one of signalling that has roles on maintain normal function of sertoli cells. Src and EGFR are some of protein that are involved in this pathway. This study focuse on formulation and characterization of nanoparticles Phyllanthus niruri, inhibition study of activation on Src and EGFR using western blot method on TM4 cell line. TM4 cell line is collected from sertoli cell of male mice. Size, polidispersity index, potential zeta of this formula are 171 nm, 0,274 and +37,82 respectively. Entrapment efficiency is 75%,. IC50 value from viability study using CCK-8 is 206 ppm. Study of activation inhibition of Src show that there are activation inhibition with significantly different occur on nanoparticle PN 500 ppm group (p<0,5). Study on EGFR show there are activation inhibition of EGFR on nanoparticle PN 7,81 ppm; 31,25 ppm; 125 ppm groups. Activation inhibition of Src and EGFR cause disruption effect on spermatogenesis and male fertility. This results may indicate there are disruption of sertoli-germ cell adhesion and release of mature spermatid. text
