Plants with high phenolic content and high antioxidant activity can also produce
high inhibition of tyrosinase activity. The purpose of this study was to determine
the inhibitory activity of the tyrosinase enzyme by methanol extract of meniran (P.
niruri L.) and to isolate secondary metabolites from the extract and fraction of
meniran (P. niruri L.) which actively inhibits the tyrosinase enzyme. Tyrosinase
inhibition activity used the 96-well microplate method in vitro with kojic acid as
positive control. Separation dan purification using chromatography techniques.
Methanol extract and n-hexane fraction had tyrosinase enzyme inhibitory activity
with IC50 of 11,05±0,16 and 2,57±0,08 mg/ml, respectively. Three bioactive
compounds have been isolated. Based on UV spectrum and HPLC overlay with
standard, isolate 1 is phyllanthin, and based on UV, 1H-NMR and 13C-NMR
spectra, isolate 3 is hypophyllanthin. Isolate 2 based on UV spectrum is considered
as compounds from the lignan group. Phyllanthin showed inhibitory activity of
enzyme tyrosinase with IC50 264,57±3,74 ?g/ml. Meniran extract and phyllanthin
were recently reported to have inhibitory acitivity against the tyrosinase enzyme
