Phytochemical studies show that this plant is a potential source of secondary metabolites which have important bioactivities. However, plants cannot be used as the main source of secondary metabolites because they have long time growth and can cause environmental damage if logging is carried out on a large scale. Therefore, alternative source of other secondary metabolites is needed. Source that can be used as an alternative are tissue culture and endophytic microorganism. There have been no reports related to the study of secondary metabolite of endophytic fungi in kecapi plants. Based on this background, a study was conducted with the aim of isolating and characterizing secondary metabolites of the endophytic fungus on Sandoricum koetjape Merr., as well as testing its bioactivity using the Docking method. Isolation of endophytic fungi was carried out by axenic culture method and a single isolate was obtained and then identified molecularly. The single isolates of the fungus were then cultured in a liquid medium in the form of Potato Dextrose Broth (PDB) dissolved in distilled water for 14 days (28°C). The cultured liquid media was then partitioned with ethyl acetate at room temperature, and the mycelia was extracted with methanol. Purification of secondary metabolites was carried out using Radial Chromatography (KR). The purified compounds were characterized using 1D- NMR (1H-NMR, 12C-NMR, and TOCSY 1D) and 2D-NMR (HSQC and HMBC). Compound 1 was then studied by docking method against eight tyrosine kinases, namely EGFR, HER2, HER4, IGF1-R, InsR, KDR, PDGFR-?, and PDGFR-?. In this study, one isolate of endophytic fungus was successfully isolated, namely Clonostacys rosea from the branch of S. koetjape Merr. Two compounds were isolated from the endophytic fungi, namely cytochalacin H (1) and 5- hydroxymethyl-2-furaldehyde (2). The molecular docking results showed that compound 1 had the best binding affinity for EGFR protein. Based on the results of this study, it can be concluded that the endophytic fungus Clonostacys rosea from the harp twig has the potential as a source of secondary metabolite diversity. text
