Peryonix excavatus is a local earthworm that abundant in tropic area like Indonesia. As shown previously, P. excavatus possess fibrinolytic activities that very useful for trombolytic agent. The aim of this research is further characterizing optimum condition of enzyme activity from this earthworm. Isolation and partially purification using fractionation of 60percent saturated amonium sulphate and gel filtration chromatrography was used to obtain the enzyme. Then, enzyme was identified by fibrinolytic assay and SDS-PAGE. In order to determine optimum condition and to analysis of purification degree, the activities of the enzyme were performed by the azocaseinolytic method. The enzyme, with molecular weight between 25-30 kDa, showed a fibrinolytic activity by diluting fibrin clots. The optimum of proteolytic activity of the enzyme was at 55degreeC and broad range pH between 6 and 12. The purified enzymes showed azocaseinolytic activity by 12percent in yield, with purity 2.5 fold higher to that of crude extract. |
