Xylanase is widely used in the paper, animal feed, food and beverage industries. Downstream processing of xylanase consists of purifying and concentrating processes. The main stage of this research is to determine the optimum parameter in every step technology. The optimum condition was achieved by UF membrane with purification fold (PF) 4,517 ± 0,809 at TMP 0.2 bar and constant CFV of 0.022 m/s using PVDF and 0.01~0.05 ?m of pore size. In precipitation of ammonium sulfate, PF 2.432 ± 1.377 was evaluated at 40% concentration and in aqueous two phase system (ATPS), the highest purity was found in 20% PEG 2000 with 22.5% MgSO4.7H2O, PF 2.367 ± 0.636. The second stage is a combination of ATPS (20% PEG 2000-22.5% MgSO4.7H2O)-UF membrane (TMP 0.2 bar). In this stage, the rate of flux obtained is 17.58 L/m².h with 28.31% (w/w) yield mass. The purity at the combination technology can be evaluated will be higher than the single-stage UF membrane technology. Finally, the addition of 5% PEG 2000 carrier at freeze drying technology was the best in increasing the mass of xylanase and even increased the enzyme purity by PF 1.501 ± 0.224. text |
