Water extract of tempuyung and madeira vine leaves as combination was proven to have antihypertensive activity, which one of the ways by inhibiting ACE activity. The usage of ACEI drugs during pregnancy is contraindicated due to its ability to cause fetal defect, including nervous system defect because of intervention during RAA system development. Unlike in human, RAA system of laboratory animal is significantly developing during late-gestational (prenatal) and continue throughout lactation (postnatal) period. This research aims to evaluate effect of tempuyung and madeira vine extract as combination given during gestational (prenatal) and lactation (postnatal) period towards development of physical appearance as well as motor and cognitive function. To evaluate the effect of extract during gestational periode, three treatment groups (n=20) were given extract at doses of 100 mg/kgBW, 400 mg/kgBW, and 1000 mg/kgBW at gestational day 6 up to 15 (G6-15) while control group was given Na-CMC 0,5% as much as 1 ml/100 g BW. Pregnant rats were terminated at gestational day 19 (G19). Number and percentage of alive, undeveloped, and dead fetus were counted and alive fetus was weighed individually. Physical examination included skeletal and visceral examination. To evaluate effect of extract during postanatal period, treatment groups (n=7) were given extract at same variance of doses as previous evaluation at gestational day 6 (G6) until postnatal day 21 (PND 21). Motor function was evaluated through reflex test, negative geotaxis test, forelimb test, traverse-narrow path test, and rope-climbing test at PND 6-21. Cognitive function was evaluated based on reward-alternation method using Y-maze at PND 25 and 26. Rat pups were sacrificed at PND 30, its brain weighed, and prepared for histology examination. Parameters were analyzed using one-way ANOVA and chi-square method (?=0,05) with IBM SPSS 28. This research concluded that consumption of water extract of tempuyung and madeira vine leaves as combination during preganancy and lactation period is contraindicated due to finding of undeveloped fetus (resorption) and some fetus with small brain at all treatment groups and decreasing of motor response at group which given extract at doses of 400 and 1000 mg/kgBW. text
