Summary:Carbamazepine is a class of first-line antiepileptic drugs commonly used to treat partial seizures, generalized tonic-clonic seizures, and trigeminal neuralgia. As a BCS (Biopharmaceutical Classification System) class II drug, carbamazepine has poor solubility. This can affect the dissolution rate and have an impact on poor bioavailability. The formation of cocrystals with suitable coformers can be an alternative to overcome these problems. However, the experimentalbased cocrystal screening method is considered less effective and efficient so that the computational method can be a profitable choice. In this study, coformers screening that have the potential to form cocrystals with carbamazepine was carried out using Mercury 2020.2.0 software from the Cambridge Structural Database (CSD) based on molecular complementarity and hydrogen bonding propensity. In addition, a literature study has been carried out regarding the possibility of carbamazepine cocrystals in improving their solubility and dissolution rate. Based on the analysis of the results of coformer screening and literature review, it was found that benzamide as the most potential coformer to increase the solubility and dissolution profile of carbamazepine.
