Summary:The increase in cases of microbial resistance to antimicrobials encourages research to find new alternative antimicrobial substance. There are many natural resources that have potential to produce secondary metabolites that have antimicrobial activity, for example endophytic fungi. This study aimed to isolate endophytic fungi from brown algae from the South Coast of West Sumatra and to test their antimicrobial activity against Mycobacterium smegmatis and Candida albicans. The steps of this study started from the isolation of endophytic fungi followed by 3-5 weeks fermentation. The fermentation products were filtered to separate the fungal mycelium culture and the media culture. The fungal mycelium culture was then extracted using maceration method with ethyl acetate as a solvent, meanwhile the media culture was extracted using the Liquid-Liquid Extraction (LLE) method with the same solvent. Extracts obtained from mycelium culture and media culture were tested for antimicrobial activity by determining the Minimum Inhibitory Concentration (MIC) using the MTT assay for Mycobacterium smegmatis. From this research, it was obtained 5 fungal isolates coded FPPS 1 to FPPS 5 and yeast isolate coded RPPS 1. The strongest antibiotic activity was shown by mycelium culture extracts FPPS 2 (50 µg/mL) and RPPS 1 (50 µg/mL) against Mycobacterium smegmatis. Based on the research results, endophytic fungi isolated from brown algae have potential as antimicrobial so that they can be considered as a new antimicrobial source.
