Kidney failure can be divided into Acute Kidney Injury (AKI) and Chronic Kidney Disease (CKD). Binahong and tempuyung have kidney protection effects. Kidney function can be determined by measuring biomarkers. Currently, various studies of specific biomarkers of AKI and CKD are developing. This study aimed to determine the effect of extracts of binahong, tempuyung, and their combination on a rat model of kidney failure and to determine specific biomarkers of AKI and CKD. Wistar rats were induced by piroxicam 3.6 mg/kgBW and gentamicin for 37 days. Gentamicin was given at a dose of 50 mg/kgBW/day on days 1 to 21 followed by a dose of 100 mg/kgBW/day on days 22 to 37. Binahong, tempuyung extract, and their combination as therapy were given for 14 days from day 38. Evaluation of kidney function was carried out by measuring serum urea and creatinine, urinalysis, kidney macroscopic, and kidney index. Biomarker data were collected through a literature review of preclinical and clinical trials of biomarkers of AKI and CKD using the search engines Pubmed and Google Scholar. Based on statistical analysis, the endpoint serum urea and creatinine levels of all test groups were significantly different with the positive control (p<0.05) and the K75-25 group had the lowest levels. Based on urinalysis, groups B100, T100, K50-50, K100-100, and K75-25 were close to negative controls. B100, T100, K50-50, and K75-25 kidneys were close to negative controls. The lowest kidney index belonged to the K25-75 group and was not significantly different from the K75-25 group (p>0.05). The K75-25 group had the best kidney function repair activity based on all parameters. Then based on a literature review, 34 biomarkers were obtained, namely 31 AKI biomarkers, 7 CKD biomarkers, and 4 of them can be used for both AKI and CKD. text |
