Summary:Binahong [Anredera cordifolia (Ten.) v. Steenis, Basellaceae] leaf is traditionally used for treating many kinds of diseases, however, the data of its chemical content is still limited. This research is aimed to study chemical content of binahong leaf and to determine identity compound of its crude drug. Phytochemical screening showed a presence of flavonoid, saponin, and steroid/triterpenoid compounds. Crude drug was macerated with ethanol and the extract was fractionated through liquid-liquid extraction. Two flavonoid compounds were isolated from ethyl acetate fraction and the isolates were purified by preparative paper chromatography and characterized using spray reagent of aluminum(III) chloride 5% and ultraviolet-visible spectrophotometry with the use of shift reagents. The isolates had ultraviolet-visible spectrum data as flavones with OH-group at positions of C 5, 7 and 4’. The flavones were an aglycone and its glycoside which could be used as identity compounds of the ethanol extract of binahong leaf. By using paper chromatography system with acetic acid 15% as mobile phase and visualized under UV light 366 nm with the use of spray reagent aluminum(III) chloride 5%, both compounds were shown as yellow fluorescent spots with Rf values of 0.25 and 0.5.
