Summary:Gout is a metabolic syndrome which characterized with recurrent periods of acute attack as a result of monosodium urate deposit in joint and cartilage. One of the goal of gout treatment is to decrease the synthesis of uric acid by inhibiting xanthine oxidase enzyme, namely allopurinol. Some plant of genus Zingiber used empirically for the treatment of gout. The purpose of this research was to prove the activity in inhibiting xanthine oxidase of Zingiber officinale Roscoe., Zingiber zerumbet (L.) J. E. Smith., Zingiber aromaticum Val., Zingiber purpureum Roxb., dan Zingiber ottensii Val.. A 100 µg/mL concentration of extract was added to the conversion reaction of xanthine to uric acid which catalized by xanthine oxidase. This method was done to the ethanol extract of dried and fresh plants. Allopurinol was used as a comparator drug. The activity of inhibiting xanthine oxidase was observed through the decreasing absorbance of uric acid. IC50 was determined for extracts that had xanthine oxidase inhibition activity value greater than 45%. There were five extract that had activity above 45% i.e., dried Zingiber officinale Roscoe. (49.9 ± 0.7%), fresh Zingiber officinale Roscoe. (45.4 ± 0.9%), fresh Zingiber ottensii Val. (55.4% ± 0.9%), fresh Zingiber zerumbet (L.) J. E. Smith. (61.2 ± 0.5%), and fresh Zingiber aromaticum Val. (46.8 ± 0.2%) with IC50 value respectively 98.3 µg/mL, 106.3 µg/mL, 94.7 µg/mL, 83.1 µg/mL, and 111.9 µg/mL. Meanwhile, 100 µg/mL allopurinol’s activity value was 89.7 ± 1% with IC50 27.5 µg/mL. The highest activity value was showed by Fresh Zingiber zerumbet (L.) J. E. Smith. rhizome extract.
