Indonesia is the 4th largest banana producer in the world based on 2018 FAO (food and agricultural organization) data. Cavendish banana (Musa acuminata) is one of the most tradeable in Indonesia. Through the tissue culture method, cultivation rates have risen significantly while shortening seed maturity duration resulting in better seed cultivation productivity. The research uses the Temporary Immersion System (TIS) tissue culture bioreactor which uses a liquid medium without continuous immersion, resulting in aeration and homogenous nutrient content. The research aimed for growth effectivity and economic feasibility analysis. The growth effectivity analyzed by biomass growth and survival rate of Cavendish banana shoot culture in liquid and TIS-RITA® bioreactor with variants of 5 and 20 minutes using Murashige & Skoog liquid medium half-strength and gibberellic acid 0 ,5 ppm planted with ex vitro method. Economic feasibility is analyzed by measuring the value added to the net present value. Based on the research of the study, banana plant biomass grown on 14 days in vitro culture in TIS Twin Flask bioreactor with immersion variation of 5 minutes and 20 minutes was increased 25% and 32%, respectively. The average growth rate of banana plant at various immersion time of 5 minutes and 20 minutes, respectively, was 0.05 cm/day and 0.121 cm/day. Also, the survivability rate of the liquid system and TIS Twin-flask bioreactor was 66,7% and 100%, respectively. Economic analysis shows great results with evidence that the NPV of the application of TIS Twin-flask bioreactor technology reaches Rp. 13,696,000.00 (payback period 2 years and 6 months) compared to Rp. 11,990,670.00 (payback period 2 years) in the thin layer culture system. text |
