Many people are aware of this impact and try to stop smoking, but it is a challenging task due to an addiction effect from cigarettes which is caused by nicotine contained in tobacco. The purpose of this study was to develop a patch formula for smoking addiction therapy with nicotine as active substance and in vivo release profile was performed in rabbits. Nicotine patch was firstly prepared from nicotine gel (0.25%) using FIPMC as the gel base (10%), then gel was dried until film layer was formed. Adhesive, polyisobutylene in n-hexane (2.5% w/v), was added to the film. Evaluation for patch include a chemical, physical, and also in vitro and in vivo release properties. Chemical evaluation will cover concentration test in patch. Physical evaluation will cover mechanical strength, thickness, and weight homogenicity test. In vitro evaluation will be done using Franz diffusion cell. In vivo evaluation will be done on rabbits dan concentration of nicotine in plasma will be checked until 24th hour. Concentration test results the %recovery from patch is 101.00+0.11% (n=6). Film evaluation results 33.08+2.41 kg mm -2 for tensile strength and 3.39+0.57% in °A elongation (n=3 each). In vitro release test using Python reticulates shed snake skin showed that the patch follow the release of Higuchi model and evaluation using rabbits show AUC0_24 3138.27 ng h for formulated patch and 2146.05 ng mt-1 h for comercial one. Formulated nicotine transdermal patch using HPMC matrix base has good physical, chemical characteristic and in vivo release with AUC0_24 which is higher than in comercial one. text
