Summary:Human interferon a2b (hIFNa2b) is known as standard therapeutic protein for hepatitis B and C treatment. However, due to rapid elimination through renal clearance, structure modification needs to be developed. Recombinant human interferon alpha 2b (rhIFNa2b) mutein form was successfully developed by site-directed mutagenesis technique. The mutein forms were developed by substituting cystein codons at positions 2 and 99 with aspartic acids. rhIFNa2b mutein C2D C99D has been proved to have better pharmacokinetic profile and stability than its native form. Therefore, this research is aimed to test its pharmacological effect in animal model. rhIFNa2b native and mutein form were obtained by overproduced the recombinant E. coil, purified by affinity chromatography column, and characterized using SDS-PAGE. Chronic hepatic inflammation in rat was induced using carbon tetrachloride given by intraperitoneal injection twice a week for four weeks. Native and mutein forms of rhIFNa2b were administrated by intramuscular injection 0,5 lig per rat while silymarin was used as a standard drug which was given orally 200 mg/kg BW. The treatment were given twice a week for six weeks. Biochemical parameter in blood serum showed that rhIFNa2b mutein C2D C99D showed better therapeutic effect in reducing level of ALT, AST, and ALP when rh11-7Not2b native proved to increase albumin level and decrease bilirubin level. Histological analysis of liver and kidney in rats treated with rhIFNa2b mutein C2D C99D group showed better improvement in organ structure compared to its native form. The results indicate that rhIFNa2b mutein C2D C99D has better anti-inflammatory effect than the native form. Further study such as protein expression and gene that involved in chronic hepatic inflammation must be conducted to get the description and rhIFNa2b mechanism of action in tested model.
