This research aimed to develop transdermal vaccine formulation consisted of nanoemulsion base, bovine serum albumin (BSA) as model immunogen protein, and artin-M as adjuvant. Artin-M is a lectin isolated from cempedak (Artocarpus integrifblia) seed. Nanoemulsion formulation was prepared using self-nanoemulsification method as the oil in water emulsion. Nanoemulsion was prepared using a mixture of virgin coconut oil (VCO), Tween 80, and PEG 400 as oil phase, surfactant, and co-surfactant, respectively. BSA and artin-M were incorporated into oil phase as solid dispersion form using PEG 20000. Evaluation of nanoemulsion consisted of BSA entrapment efficiency, hemagglutination activity of artin-M, and physicochemical stability test including the measurement of globule size, polydispersity index, viscosity, zeta potential, and pH. The optimum formula of nanoemulsion comprised of 3% VCO, 16% Tween 80, 8% PEG 400, 5 mg/g solid dispersion of BSA, and 50 ug/g solid dispersion of artin-M. The average of globule diameter was 42.7 ± 0.9 nm and polydispersity index was 0.321 ± 0.01. BSA entrapment efficiency in oil phase was 98.6 + 0.15%. Hemagglutination activity test showed that artin-M in the nanoemulsion had activity up to 3.12 ppm. The stability test showed that nanoemulsion was relatively stable during 24 days of storage. |
