Exposure to free radicals can be prevented by the presence of antioxidants. Basically, our bodies are producing natural antioxidants, however the amount of antioxidants are not enough to neutralize free radicals in the body. Therefore, it takes an extra antioxidants from outside the body. Cinnamon is a plant which contains several compounds such as phenol, flavonoid, and tannin that have antioxidant activity. This research aims to determine the 1050 of ethanol extract of cinnamon bark and to measure the antioxidant activity of emulgel preparation (in vitro). For in vivo test, primary and okular irritation in rabbit were performed. Crude drug was extracted by reflux method using ethanol 96%. Thin layer chromatographic monitoring was done by using solvent systems of toluene-ethyl acetate (97:3), measurement of DPPH (2.2-difenil-1-pikrilhidrazil) scavenging activity and determination of IC50 with ascorbic acid as positive standard were also performed. Concentrated extract made in an emulgel formulation which previously has been optimized. Ernulgel of ethanol extract of cinnamon bark used virgin coconut oil (VCO) as phase base oil. Stability test preparations include organoleptic observations (color, odor, phase separation, microbial growth), freeze and thaw test, determination of pH and viscosity, accelerated stability test using climatic chamber (40°C), and measurement of DPPH scavenging activity. primary and okular irritation in rabbit has been done. Ethanol extract of cinnamon bark contains alkaloid, flavonoid, tannin, steroid/triterpenoid, and saponin. Ethanol extract of cinnamon bark (density extract 1% 1.0048 pg/mL) provide a high DPPH scavenging activity with the value of IC50 is 8.36 tg/mL compared to ascorbic acid (1050 8.85 p..g/mL). An emulgel of ethanol extract of cinnamon bark that consists of VCO 20%, glycerin 10%, sodium lauryl sulfate 0.7%, cetostearyl alcohol 6.3%, PVP 3%, and ethanol extract of cinnamon bark 0.84% showed a relatively good physical stability. Percentage of DPPH scavenging activity this emulgel during observations was in range of 40.81 — 50.67%. Emulgel didn't irritate the skin and eyes of albino rabbits. text
