Folic acid is one example of the vitamins. As micronut•ient the folic acid in food is relatively small. Folic acid assay in vegetables had been developed by many methods and extraction processes. It is relatively difficult to analyze because this vitamin is low, unstable and appears in diverse forms in food. This research aimed to determine the content of folic acid in spinach, broccoli and multivitamin of vegetables extract by microbiological assay from folate-dependent growth response of Lactobacillus cctsei BTCC B633. The folic acid reference standard assay was optimized by selecting acetic buffer pH 4.5, citric buffer pH 5.0 and sodium hydroxide (NaOH) 0.01 M as the solvent. Folic acid in tablet was solved with each solvent. The calibration curves were prepared. The best regression of the curves was chosen from the solvent used. Spinach and broccoli were dried and prepared to be a crude drug. They were extracted by maceration and reflux method, each crude drug was extracted with water and ethanol 96% (v/v). Microbiological assay using Lactobacillus easel and the growth of bacteria were compared by measuring the turbidity of different samples after incubation period. The absorbance value was measured with spectrophotometer UV-VIS at 2., 650 nm. The content of folic acid in samples was determined by extrapolation from the standard curve. NaOH 0.01 M was chosen to be used for calibration curve with regression value, 0.995. The amount of folic acid in spinach and broccoli crude drug was found at the value 13.08 qg/g and 10.81, sample respectively. The highest amount of folic acid was found in the spinach ethanol extract that extracted with reflux method in a value 35.48
/g extract. The amount of folic acid in multivitamin from two brands of vegetables extracts were 46.74 dan 56.44 ugig sample respectively. Microbiological assay using Lactobacillus easel BTCC B633 can be used to determine folic acid in spinach, broccoli, and multivitamin from vegetables extract with high selectivity and acceptable accusation and precision based on recovery percentage of calibration curve in a range 95-105%.
