Complementary food such as infant formula is the first choice as supplementary nutrition for infants. Most of infant milk formulas in market contains docosahexaenoic acid (DHA). As an unsaturated fatty acid, DHA act as an essential substance for growth and development of cerebral tissue in the brain. Most of infant milk formulas are in instant powder form which in its preparation of ready-to-drink need warm water for dilutation. This research purpose was to validate the method of determination DHA content of the ready-to-drink infant formula using the variance treatments of temperature. On other hand, this work also expected to determine the optimum temperature for preparation of infant formula by observe if there was effect of heating in the preparation. The sample preparation steps were started with extraction, esterification, up to the samples were measured by gas chromatography elution programmed system at initial temperature 150°C for 0.5 minutes, then elevated up to 248°C at flow rate 9 °C/minutes, OV-1 capillary column (25 m x 0,53 mm), nitrogen gas, FID at 280°C, and the injector at 250°C. The DHA concentration prepared at 30°C was 0.391 ± 0.007 mg/g sample, prepared at 60°C was 0,400 ± 0.023 mg/g sample, and prepared at 90°C was 0.401 ± 0.017 mg/g sample. The temperature treatments showed no significant difference, so that, there was no effect of temperature on the preparation of the ready-to-drink infant milk formula. There was closed of DHA concentration measured from the sample with DHA concentration from product labels i.e 94.52% w/w. text
