Summary:Sweetener is one of food additives for daily use and industrial. Artificial sweeteners such as, aspartam, acesulfame-K, saccharin, sodium saccharin, calcium saccharin, potassium saccharin, cyclamic acid, sodium cyclamate, calcium cyclamate, sucralose, and neotame are permitted to be used in Indonesia. Excessive use of artificial sweeteners can lead to certain health effects, therefore a safety evaluation of artificial sweeteners is needed. Toxicity prediction is one of the method that capable to evaluate the safety of the artificial sweeteners. This study was aimed to predict the toxicity of artificial sweeteners that are allowed in Indonesia using ADMET PredictorTM software. Modeling and geometry optimization was performed by HyperchemTM software with Ah initio method. Moleculs of artificial sweeteners that have been optimized, were calculated using ADMET PredictorTM. The research resulted prediction toxicity of eleven artificial sweeteners. Aspartam showed no toxicity whereas cyclamic acid was hepatotoxic and induced cutaneous and respiratory allergic reactions. Calcium cyclamate caused toxicity in the androgen receptor, induced cutaneous allergic reactions, and caused reproductive toxicity. Saccharin showed hepatotoxicity and induced cutaneous allergic reactions. Sucralose caused toxicity in the androgen receptor, hepatotoxic, and caused chromosome aberration and inutagenic. However, the toxicity of acesulfame-K, sodium cyclamate, saccharin calcium, potassium saccharin, sodium saccharin, and neotame can not be determined due to its lack of accuracy in analysis parameters.
