Recently was reported a research that aims to lower the dose and toxicity of chloramphenicol and oxytetracycline via combined these antibiotics and complexed them with Fe (II), Co (II) and Ni (II). Ni(CHL)(OTC)C12 complex was reported had higher activity level than the original compound, and the lowest toxicity level compared to the others. Research has developed methods to produce Ni(CHL)(OTC)C12 complex and characterized the complex with a solid characterization instruments, such as Fourier-Transform Infrared Spectroscopy (FTIR), Differential Thermal Analysis (DTA), polarization microscopy, and Powder X-ray Difractometri (PXRD). Ni(CHL)(OTC)C12 complex was composed using three methods : co-grinding method, reflux method, and pH adjustment method. The crystalline solids obtained were analyzed and characterized using TLC, Fourier-Transform Infrared Spectroscopy (FTIR). Differential Thermal Analysis (DTA), polarization microscopy, and X-ray Powder Difractometri (PXRD). The most stable crystalline complex and the highest yield were obtained from pH adjustment methods that produce 24,31 % complex rendemen. The complex crystals has yellowish green color, slightly more polar than their raw material which are chlorampenicol and oxytetracycline, and the results of three-dimensional TLC and two-dimensional TLC indicate that the obtained complexes are mostly pure. IR spectrum from the complex indicates that the acidoxyl groups shifted from 1631cni I to 1523 cm', nitrophenyl shifted from 1523 cm-' to 1349 cm-' and a new peak appeared on 689 cm-' showed the interaction with the metal. The DTA thermogram showed endhotermic curve that indicated melting range temperatures at 133° C and 140° C. Observation with polarizator microscopy showed that Ni(CHL)(OTC)C12 is a rod-shaped crystal. Diffactogram complex showed unique peaks at 20 = 33,86°, 39,94° and 44,66°. Base on data and analysis results, it can be concluded that the best method for Ni (CHL) (OTC) complexation is the adjusting pH methods. The chrystal is quite stable. showing a typical FTIR spectra in the wave number 1685, 1349, dan 689 cm'', melting point 133°C and 140°C, and rod-shaped crystals .Internally is a new crystal structure that has not been reported previously, with typical diffraction peaks at an angle 20 = 33.86°; 39.94°and 44.66°, and it's structure is potential to be determinedby single crystal X-ray diffraction.
