Therefore, acidic chitinase is required for that purpose Chromobacterium violaceum is bacterium isolated from Indralaya swamp area located in South Sumatera and is expected to produce acidic chitinase. C. violaceum was confirmed bt Gram staining and Polymerase Chain Reaction (PCR) 16s rDNA as well as to be chitinase producer using qualitative chitinase activity assay in Nutrient Agar medium containing colloid chitin. The nucleotides predicted to contain a fragment gene encoding the first 197 amino acids of chitinase has been cloned into pGEM-T in E. coli JM 109 based data obtained from migration analysis, PCR analysis, and cleavage analysis by restriction enzyme. <br /> text |
