Summary:Indonesia have many kinds of tropical fruit, but the rind and the peel are usually wasted, some of them are watermelon and dragon fruit. Literature showed watermelon rind [Citrullus lanatus (Thunberg) Matsurn. & Nakai] and dragon fruit peel [Hylocereus undatus Britt. & Rose] contained several compounds which is expected to have immunomodulatory property. Therefore, this research was carried out to determine the immunomodulatory activity of ethanol extract of watermelon rind and dragon fruit peel. The extraction was performed by maceration-percolation method using ethanol 96%. Immunomodulatory activity was studied in Swiss Webster mice by determination of phagocytic index of reticuloendothelium system through carbon clearance test, antibody and IFN-7 titer. The result of carbon clearance test showed that ethanol extracts of dragon fruit at 150 and 300 mg/kg bw have immunostimulant properties with phagocytic index greater than one. Both doses of ethanol extract of watermelon rind tested doses increased the liver and spleen index significantly (p<0.05) compared with control group. However both doses of ethanol extract of dragon fruit peel increase: the liver index significantly (p<0.05). Both doses of ethanol extracts of watermelon rind and dragon fruit peel increased the primary antibody titer. Secondary antibody titer increased after ethanol extract of watermelon rind at 160 mg/kg bw, and both doses of ethanol extract of dragon fruit peel. Ethanol extract of watermelon rind at 160 mg/kg bw reduced the production of IFNI, significantly (p<0.05). Based on this research, the ethanol extract of dragon fruit at 150 and 300 mg/kg bw and ethanol extracts of watermelon rind at 80 and 160 mg/kg bw have weak immunostimulation activity with ethanolic extract of watermelon rind at 160 mg/kg bw reduced the production of IFN-7 significantly (p<0.05).
