The Crude drug of H. roxburghii Bl. rhizome was extracted by water steam distillation to obtain the essential oils. Distillation residue was dried by freeze dryer, and then it was extracted by reflux method using ethanol as a solvent. The crude drug and ethanol extract of distillation residue from H. roxburghii Bl. rhizome contained flavonoid, phenol, quinone, and steroid/triterpenoid. Antibacterial activity of essential oil and ethanol extract of distillation residue were tested by microdilution method using Mueller Hinton Broth (MHB) on 96-microwell-round-bottomed plates against B. subtilis (ATCC 6633), S. aureus (ATCC 6538), E. coli (ATCC 8939), and P. aeruginosa (ATCC 9027). Minimum Inhibitory Concentration (MIC) was determined based on the smallest concentration where the microwells free from precipitate sedimentation at the bottom of the well compared with control. Essential oil showed antibacterial activity against B. subtilis (ATCC 6633), S. aureus (ATCC 6538), E. coli (ATCC 8939), dan P. aeruginosa (ATCC 9027) with MIC respectively 1.76; 1.76; 7.03; and 1.76 ug/mL. Ethanol extract of distillation residue showed antibacterial activity against B. subtilis (ATCC 6633), S. aureus (ATCC 6538), E. coli (ATCC 8939), and P. aeruginosa (ATCC 9027) with MIC respectively 128; 256; >256; and 256 ug/mL. Bioautography test results showed that the essential oils on GF254 TLC chromatogram with mobile phase system of toluene-acetone (9.5:0.5) showed antibacterial activity at Rf 0.3; 0.6; and 0.76, while the ethanol extract of distillation residue with mobile phase system of chloroform-methanol-acetic acid (9:1:2 drops) at Rf 0.5 and 0.78. text |
