Some research have shown that binahong extract has several activities such as antioxidant, anti-inflamation, and antihiperuricemia. There has been some research that shows binahong contains flavonoid compounds. Flavonoid is a group of natural compound which has been reported to have antioxidant activity, hepatoprotector, and anti inflamation. The aim of this research is to isolate and characterize the flavonoid compound in binahong extract. One kilogram of binahong leaves sample was extracted by maceration method using ethanol 96 %. The extract was concentrated by vacuum rotary evaporator and monitored using thin layer chromatography (TLC) with silica gel GF254 as stationary phase, chloroform:methanol (8:2) as mobile phase, and citroboric acid as visualizing agent Fractionation was done by modified Charaux-Paris method, which was liquid-liquid extraction using three different solvent with increasing polarity, n-hexane, ethyl acetate, and n-butanol. Fractions monitoring was done by using TLC with chloroform:methanol (8:2) as mobile phase. The butanol fraction was purified using preparative paper chromatography with water as stationary phase and acetic acid 10% as mobile phase. Purity test was done by TLC single development using three different polarity mobile phase. Isolates were characterized using spectrophotometer ultraviolet-visible with shifting reagents and 2 dimensional paper chromatography. Flavonoid, saponin, steroid/triterpenoid, phenol, quinone were detected in both binahong sample and extract. There were three isolates obtained from butanol fraction of binahong extract. Isolate 1 is a flavone that has 7-OH and predicted having one sugar (monoglycoside) attached to 0- on C-5. Isolate 2 is a flavone which has —OH on C-7 and predicted having 5-OH without —OH on C-4. Isolate 3 is a fiavone that has 7-OH and o-diOH on ring B, and predicted having sugar attached to C-5. |
