Summary:Over time, the prevalence of liver disease has been developing with the severity and risk of death increase. Highly liver's performance and its detoxificating function influence various kind of liver diseases arise. Liver disease can be caused by some factors such as unhealthy life style, stress, and exposure to toxic compounds that induce the release of free radical compounds. These compounds will disrupt metabolism function and cause damage to the liver cells. Sweet potatoes are a source of food that has been known around the world, especially Indonesia. In addition, the recent researches proved that sweet potato cultivar Ayamurasaki has antiproliferative efficacy of cancer cells, antihiperglicemic, antimicrobial, antihypertensive, and in vitro antioxidative activity. This study is aimed to test hepatoprotective activity from the ethanol extract of sweet potato tuber and its juice product against compounds-induced liver injury in mice. The assessment of liver function including enzyme activities of AST, ALT, bilirubin level, organ index, and liver histopathology was determined after oral administration of sweet potato ethanol extract 100 mg/kg bw and 200 mg/kg bw; sweet potato juice 5 mL/kg bw and 10 mL/kg bw on each group, and silymarin 200 mg/kg bw as compared group for 4 weeks and intraperitonial administration of carbon tetrachloride-induced hepatotoxic 0.4 mL/kg bw once a week since 2" to 4th week. The final measurement results were compared with the initial results before treatment. The control group showed an increase in activity of AST, ALT, and bilirubin levels significantly to the initial state and indicate the occurrence of liver damage. Ethanol extract of sweet potato 100 and 200 mg/kg bw, sweet potato juice 5 and 10 mL/kg bw did not showed change on activity of AST, ALT, and bilirubin levels significantly to the initial state and had better hepatoprotective activity than silymarin. Ethanol extract 200 mg/kg bw and sweet potato juice 5 mL/kg bw had highest decreasing percentage of AST activity and bilirubin levels, also the lowest increasing percentage of ALT activity, significantly different from the control group. Microscopy examination of liver section showed that ethanol extract 100 and 200 mg/kg bw groups have well-formed hepatocytes and almost normal liver architecture. with mild degree of liver damage. Sweet potato cultivar Ayamurasaki is recommended as a good hepatoprotector agent which is able to minimize liver damage and stimulates cell regeneration. The ethanol extract at dose 200 mg/kg bw and juice at dose 5 mL/kg bw have the most effective hepatoprotective activity. Its activity is suspected that anthocyanin in these sweet potato contributes as an antioxidant compounds that counteract free radical reactions of carbon tetrachloride.
