Normal value for blood pressure in adults is <120 mmHg for systole and <80 mmHg for diastole. Based on several studies, there were increased levels of cardiac collagen in hypertensive patients. Increased levels of cardiac collagen could increase the heart stiffness which caused abnormal heart pump. Based on studies, antihypertension drug administration such as captopril and spironolactone could prevent the increasing of cardiac collagen levels. This study aimed to compare the effects of captopril and spironolactone on cardiac collagen levels. The rats were divided into 6 groups. Negative control group, positive control group which are given NaC1 3.75 g/kg bw, and tested groups which are given NaCI 3.75 g/kg bw since the beginning of the study until the end of week 5 and antihypertensive drugs since week 3 until week 5. Antihypertensive drugs were captopril (50 mg/kg bw and 100 mg/kg bw) and spironolactone (20 mg/kg bw and 80 mg/kg bw). Frequency, amplitude, systolic and diastolic blood pressure were measured every week. At the end of study, the rats were sacrificed and their hearts were isolated. The heart was weighed and heart histology was made. Masson's trichrome stain used to stain cardiac collagen. The heart histology was observed under a microscope and then analyzed using image analysis software ImageJ to determine cardiac collagen levels. Captopril and spironolactone at all doses can lower amplitude, systolic, and diastolic blood pressure. Significant difference in systolic and diastolic blood pressure (p<0.05) occurred at week 4 and week 5. Significant difference in amplitude (p<0.05) occurred at week 3 until week 5. Captopril and spironolactone also can prevent the increasing of cardiac index and cardiac collagen levels. Significant difference in cardiac index and cardiac collagen levels (p<0.05) only shown by rats given captopril 100 mg/kg bw. Captopril 100 mg/kg bw had better effects than captopril 50 mg/kg bw and spironolactone 20 mg/kg bw and 80 mg/kg bw because captopril 100 mg/kg bw could lower amplitude, systolic, and diastolic blood pressure. Captopril 100 mg/kg bw also provided significant differences (p<0.05) in cardiac index and cardiac collagen levels. text
