STUDY OF SILK FIBROIN AND SPIDROIN BASED SCAFFOLD FABRICATING FOR ARTIFICIAL SKIN EXTRACELLULAR MATRIX USING ELECTROSPINNING PROCESS

In the recovery of skin injuries, fibrous-scaffold is used as a medium for cell regeneration. Fibrous-scaffold is made using silk fibroin because it has superior biocompatibility, biodegradation, and bioactivity. Moreover, it is necessary to add spidroin which has the arg-gly-asp sequences (RGD)...

Fibrous-scaffold is made using silk fibroin because it has superior biocompatibility, biodegradation, and bioactivity. Moreover, it is necessary to add spidroin which has the arg-gly-asp sequences (RGD) to enhance cell attachment and proliferation. In order to support the recovery of skin injuries, processing with electrospinning is used to obtain fibrous-scaffold with the suitable properties. Hence, a study was made to predict the feasibility of fabrication of fibrous-scaffold based on a mixture of silk fibroin and spidroin by electrospinning using literature study as a method. In this study it was found that the fabrication of silk fibroin based fibrousscaffold using electrospinning has been extensively carried out. On the contrary, the fabrication of spidroin based fibrous-scaffold using electrospinning is not frequently done. In order to be mixed, the compatibility of silk fibroin and spidroin needs to be improved by using cross-linking methods such as using enzymes or photo-cureable cross-linking agents. Therefore, a mixture of silk fibroin and spidroin can then be electrospun using parameters: silk fibroin in the form of filament; composition of silk fibroin-spidroin less than 10%; concentration of formic acid dissolution of more than 92%; cross-linking reaction using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC); electrospinning process with a maximum K value of 3.2; isourea purification; and methanol treatment. text
