Summary:Free radicals can cause degenerative diseases, that can be neutralized by antioxidants. Green honey deli water apple is one of the Indonesian plants that contain antioxidants. This research is aimed to determine the antioxidant activity of green honey deli water apple leaves, branches, fruits through DPPH (2,2-diphenyl-1-picrylhydrazyl) and CUPRAC (Cupric ion reducing antioxidant capacity) methods, total phenolic content (TPC) and total flavonoid content (TFC), analyze the correlation between TPC and TFC with the antioxidant activity, analyze the correlation between the two methods, and determine the content of several flavonoid compounds in ethanol extract of green honey deli water apple fruit. The determination of antioxidant activities through DPPH and CUPRAC methods, TPC, and TFC were done by spectrophotometry UV-Vis. Correlation analysis was explored using high performance liquid chromatography. Antioxidant activities of leaves, branches, and fruits extracts of green honey deli water apple according to DPPH and CUPRAC were 3.97? 354.96 mg AAE/g; 10.46?222.51 mg AAE/g respectively. Ethanol leaves extracts had highest TPC (68.14 ± 1.69 g GAE/100 g) and ethyl acetate leaves extracts gave the highest TFC (18.65 ± 1.04 g QE/100 g). TPC and TFC had significant and positive correlation with the antioxidant activity. Correlation of antioxidant activities using DPPH and CUPRAC methods was positive and significant, thus the two methods showed linear results. Phenolic and flavonoids compounds had great contribution on antioxidant activities of green honey deli water apple extract using DPPH and CUPRAC methods. The ethanol fruits extract contained quercetin and kaempferol, and the highest content was stated by kaempferol (0.39%).
