Previous studies showed that the phenolic compounds and essential oil content in cinnamon have tyrosinase inhibitory activity. This study aimed to determine the potential of ethanol extract and cinnamon bark fraction (Cinnamomum burmannii (Nees & T. Nees) Blume) as inhibitors of the tyrosinase enzyme and isolate the active compound. Simplicia powder was extracted by the maceration method using 96% ethanol as solvent and monitored by thin-layer chromatography. Fractionation was carried out by liquid-liquid extraction method using n-hexane, ethyl acetate, and ethanol-water as solvents. Quantitative activity tests on extracts and fractions were carried out. The absorbance data processed into the value of % inhibition at each concentration and IC50 was determined by linear regression between logs concentration and % inhibition. The IC50 values of extract samples, n-hexane fraction, ethyl acetate fraction, and water-ethanol fraction were 9.442,41 ± 61,37 ; 728,95 ± 27,09; 1.173, 42 ± 45,15; and >10.000 µg/mL. Using bioautography thin layer chromatography method, white spots on a gray background indicated enzyme inhibitory activity. Subfractionation was carried out by classical column chromatography, and selected subfractions were purified using the preparative thin layer chromatography method. The purity test was carried out using single expansion thin layer chromatography with three mobile phases and twodimensional thin layer chromatography. The purity test results obtained isolates W, isolates X, isolates Y, and isolates Z. The isolates were then characterized using spesific spray reagent. The ethanolic extract of cinnamon bark contains compounds with phenol, flavonoid, coumarin, and triterpenoid groups. The nhexane fraction gave more potent inhibitory activity to the tyrosinase enzyme than the ethanol extract, ethyl acetate fraction, and ethanol-air fraction. From the n- hexane-fraction, isolates W, X, Y, and Z were obtained, the result showed triterpenoid group compounds presence and qualitative inhibitory activity of the tyrosinase enzyme. text
