Summary:Diabetes mellitus is a metabolic disorder that has become a common problem globally. One of the approaches in diabetes treatment is by inhibiting ?-glucosidase activity. This study aimed to determine inhibition activity of ?-glucosidase of Syzygium myrtifolium Walp. leaves from four different regions. Extraction was carried out by maceration method using ethanol 96%. Inhibition activity of ?- glucosidase was determined by a microplate reader, using acarbose as a positive control. The most potent extract was fractionated by liquid-liquid extraction using n-hexane, ethyl acetate, and water. The most potent ethanol extract of S. myrtifolium originated from Kabupaten Bandung with an IC50 value of 0.614 ± 0.004 ?g/mL. The ?-glucosidase IC50 value from ethyl acetate and n-hexane fraction was found to be 0.776 ± 0.020 and 3.019 ± 0.083 ?g/mL respectively. IC50 value of acarbose was 51.551 ± 0.791 ?g/mL. The ethanol extract was found more potent in inhibiting ?-glucosidase activity than ethyl acetate and n-hexane fraction. This study showed S. myrtifolium extract and fraction had higher inhibition activity than acarbose as a positive control. Thus, it clearly indicates the potential of S. myrtifolium extract, ethyl acetate fraction, and n-hexane fraction originating from Kabupaten Bandung as an ?-glucosidase inhibitor.
