Diabetes melitus can cause complications, such as diabetic neuropathy, which is a nerve disorder caused by diabetes and is characterized by tingling, pain, or numbness. The other complications are retinopathy and cardiovascular disease. The ?-glukosidase enzyme plays a role in the breakdown of carbohydrates into glucose in the digestive tract so that it can cause an increase in blood glucose levels. One potential plant source that becomes a candidate as an ?-glucosidase enzyme inhibitor is the meniran hijau herb (Phyllanthus niruri L.). This research aimed to determine inhibition activity of ?-glucosidase of extracts and fractions of Phyllanthus niruri L. through the determination of IC50 values using Microplate Reader. Ethanol extract of meniran hijau herb from four different areas with a concentration of 2 µg/mL had % inhibition values in the range of 34,08-57,23% and a concentration of 4 µg/mL in the range of 50,24-67,09%. Sample from Bandar Lampung has a highest % inhibition value. The IC50 values of ethanol extract, ethyl acetate fraction, and n-hexane fraction from samples from Bandar Lampung City were in the range of 0.86 ± 2.59-3.24 ± 4.78 µg/mL. Rutin became marker compound in meniran hijau herb with content in ethanol extract dan ethyl acetate fraction of Phyllanthus niruri L. from Bandar Lampung was 0,0048% and 0,075%, respectively. The extracts and fractions of the meniran hijau herb had a strong ?-glucosidase enzyme inhibitory activity than the positive control of acarbose. text
