This substance has low solubility in water and tend to be unstable. Therefore, a delivery system is needed to improve the ability of TA to penetrate through stratum corneum to the epidermis and increase the stability of TA, namely in the form of Nano Lipid Carrier (NLC). The objective of this study is to formulate TA in the form of NLC, then incorporated into a cream dosage form. Characterization and evaluation carried out include organoleptics, morphology, particle size, polydispersity index, zeta potential, diffusion ability, antioxidant potency, stability, and safety. The optimation of NLC formula was conducted by trial the variable such as concentration of lipid, surfactant, actives, and the method. Based on optimation, the best formula obtained from the study was consist of the combination of surfactant of Tegocare: Poloxamer 188: Tween 80 in ratio of 1:1:1 and lipid phases consist of isopropyl myristate, cethyl alcohol and TA 2%. This NLC has efficiency of entrapment of 87,14% ± 0,63; particle size of 170,3 nm ± 3,07 and index polydispersity of 0,251 ± 0,01. This NLC also has a spherical form and zeta potential value of - 32,64 ± 4,23; also stable during storage for 42 days, both in room temperature and 40 ? C. Then, NLC TA and TA (without NLC) are incorporated into cream base that consist of stearic acid and olive oil. The stability test of the cream showed that the organoleptic, pH, and viscosity are stable during storage. The potency of antioxidant and assay revealed that there is a significant different between NLC, cream NLC TA and cream TA during storage for 56 days, in the room temperature and 40 ? C. The diffusion test exhibited that the penetration of cream NLC TA was 3 times better than cream TA, which are 70,42 % ± 5,32 and 22,60 % ± 4,11 after 7 hours, respectively. The cream NLC TA was followed the Higuchi Diffusion model. The irritation test showed that cream NLC TA did not irritate the skin and eye of the rabbits. text |
