To overcome this problems, curcumin nanoemulsion and nanosuspension gel were developed for transdermal route to improve curcumin permeability. Curcumin nanoemulsions were prepared by self/nanoemulsification method. Glyceryl monooleate (GMO), cremophor RH40 and polyethylene glycol 400 (PEG 400) were used to formulate nanoemulsion. Curcumin nanosuspensions were prepared by top/downmethod using ultrasonicator with various stabilizers, i.e. polyvinil pirrolidon, polyvinil alcohol, tween 80, sodium lauryl sulfate, and carboxymethyl cellulose sodium. Evaluation of curcumin nanoemulsion and nanosuspension was carried out including to particle size analysis, polydispersity index, zeta potential, physical stability and morphology (TEM and microscopy). In addition, curcumin and nanocurcumin were incorporated into gel using 5% viscolam as gelling agent. Evaluation of gel was done including organoleptic, pH, viscosity, drug content stability, and diffusion test using vertical diffusion cell. Stable nanoemulsion which was spontaneously formed has loading capacity of 350 mg curcumin per 10 gram mixture of oil phase with mean globule diameter of 85,0 + 1,5 nm, polydispersity index of 0,18 + 0,02, and zeta potential of /5,9 + 0,3 mV. Relatively stable nanosuspensions was generated using polyvinil alcohol as stabilizer and has mean particle diameter of 814,9 + 11,5 nm, polydispersity index of 0,37 + 0,03, and zeta potential of /6,7 + 0,4 mV. Stability test showed that nanocurcumin gel was more stable compared with curcumin gel, especially in term of drug content. Diffusion test resulted in significant increasing of curcumin permeation flux (P < 0,05) of nanoemulsion gel (1,698 µg/(cm 2 .hour)) and nanosuspension gel (1,146 µg/(cm 2 .hour)) compared with conventional curcumin gel (0,835 µg/(cm 2 /hour)). The results demonstrated successful development of curcumin nanoemulsion and nanosuspension gel which are physicochemically better than curcumin gel. text |
