IMMOBILIZATION OF PPO (POLYPHENOL OXIDASE) ENZYME WITH SOL-GEL ON CARBON PASTE ELECTRODE SURFACE FOR VOLTAMMETRY DETERMINATION OF BPA (BISPHENOL A) AND BPB (BISPHENOL B)

BPA (Bisphenol A) and BPB (Bisphenol B) is one of basic components in manufacture of polycarbonate plastic which often used as food or beverage containers. Some researches show that BPA molecules can be released from the plastic in inadequate storage conditions so they can be ingested by human. Mean...

Some researches show that BPA molecules can be released from the plastic in inadequate storage conditions so they can be ingested by human. Meanwhile, presence of excessive BPA in human body is harmful to health because BPA is classified as EDC (endocrine disruptors) can interfere with the activities of the endocrine system by imitating estrogen functions in the body. Therefore, determination of BPA in food or beverage containers is important to do. In this research, Voltammetry analysis method have been developed using carbon paste electrode (CPE) modified with polyphenol Oxidase (PPO) for determination of BPA compound. Modification of CPE surface was conducted by enzyme immobilized process in a solution containing BPA standard in 0,1 M phosphate buffer solution as supporting electrolyte. Measurement of optimum composition, pH of analyte solution and effect scan rate have been investigated. Analytical measurement results using square wave voltammetry (SWV) show that the modification can increase sensitivity and stability of electrode. The calibration curve provides a linear range of biosensor was from 2 to 10 ?M for BPA and BPB with limit of detection (LOD) of 0,520 ?M dan 0,720 ?M. respectively. Measurement BPA in reusable drinking bottle water samples showed accurate results and were not significantly different compared to HPLC. text
