One method that can be used is micropropagation of stevia shoot culture in vitro in the TIS-RITA® bioreactor, and to increase its metabolites, we can use methyl jasmonate (MeJa) 10 ?M as elicitor. This study aimed to determine the effect of the etelicitation of methyl jasmonate on the growth and secondary metabolite content of stevia shoots. The results of this study showed that there was an increase in the number of shoots by 275 ± 2.7% and RGR of 0.127 ± 0.01 g/gday in methyl jasmonate elicited culture, while in the treatment without elicitation, an increase in the number of shoots was obtained by 232 ± 9.7% and RGR of 0.122 ± 0.03 g/gday. The metabolites obtained in the form of stevioside and rebaudioside-A were 3.3 and 3.1 times greater, respectively than the treatment without elicitation. The analysis performed on the cultivation medium showed a decrease in the levels of sucrose, pH, and conductivity of the medium in both treatments compared to the initial conditions. Accumulation of actual biomass is greater than the theoretical gain, which is 2.2 times greater in the elicited treatment and 2.7 times greater in the untreated treatment. Cultivation of Stevia rebaudiana in the TIS-RITA® bioreactor by immersing 30 minutes at 6 hour intervals stimulated by 10 ?M methyl jasmonate was more effective for increasing growth and accumulation of metabolites in the form of stevioside and rebaudioside-A in Stevia rebaudiana leaves. text |
