The transmission of this dengue fever virus comes from the bite of the female aedes aegyepti mosquito. One of the efforts to overcome this DHF outbreak is with an early detection of this virus, which generally uses the NS1 Dengue Virus antigen as a detected protein. In this study, an electrochemical biosensor based on MOF (Metalorganic Framework) HKUST-1 material was developed which was modified with the Trianethanolamine modulator. HKUST-1 is MOF material formed from Cu (Copper) and H3BTC (trimesic acid) organic ligands. Material synthesis uses the co-pecipitation method with water solvents, and TEOA is used as a pH regulator and structure of HKUST-1 Addition of Triathanolamine varies of concentrations of 10 WT%, 20 WT%and 30 WT%. The sample characterization uses X-ray diffraction (XRD), scanning Elecktron Microscope (SEM), Fourier Transform Infra-Red (FTIR). The process of immobilizing monoclonal antibodies is carried out with EDC-NHS Coupling with HKust-1/Teoa 30%matrix material. Electrochemical immunosensor performance is evaluated using the Cyclic Voltametry (CV) method, and Differential Pulse Voltametry (DPV). All tests are carried out with PalmSENS Instruments on 3 mm K4[Fe (CN) 6] in PBS 0.01 M. Validation of the success of immobilization of each bionial is observed by testing DPV. The result of characterization shows that the addition of teoa changes the process of growth, shape and size of the HKUST-1 crystal produced. The characterization of FTIR shows the effect of adding teoa to increase the intensity of the vibration of the N-H and C = O group which acts as a site provider for the process of immobilizing antibodies. Further testing shows that the modified electrode succeeded in detecting the presence of NS-1 DENV-3 antigens as its target. text |
