Palm oil is widely used in the food sector and contains phytonutrients that are good for health. Currently, almost all of the CPO processes are through the “thermochemical platform” route which can be developed to a greater extent through the “biochemical platform” route that is more environmentally friendly. Aquaeous enzyme-assisted oil extraction is one of the “biochemical platforms” route for oil extraction. The purpose of this study was to produce and determine the activity of crude enzymes by means of semi solid fermentation of mesokarp as a substrate using Aspergillus niger and and to evaluate the effectivenes these crude enzymes on the Aquaeous enzyme-assisted oil extraction process on the recovery of crude palm oil (CPO) as well as to determine the content of free fatty acids and the content and activity of antioxidants contained in the CPO namely: carotene, phenolic, alpha tocopherol. Fermentation was carried out with variations in time period of 3,5, and 7 days. The activity of cellulase and pektinase on crude enzymes on feremenation period of 3, 5, and 7 days respectively were 0.21, 0.27, and 0.29 U/mL and 0, 48, 0.51, and 0.50 U/mL. In crude enzymes, the activity of the tanase enzyme was detected only on fermentation time period of 5 days, which was 0.45 U/mL. The CPO yields respectively were 73.60, 79.61, and 81.12% (g of CPO/g of total fat). Using the crude enzymes produced at the fermentation time period of 3, 5 and 7 days, the recovery CPO contains respectively: free fatty acids of 4.3%, 4.8%, and 5%, , beta-carotene content of 659 ppm, 674 ppm, and 740 ppm, total phenol content of 152, 219, and 233 mg GAE/kg. extract, tocopherol content of 45.98, 73.72 and 100.89 mg/kg oil, and antioxidant activity of 767.94, 810.4, and 816.98 mg Trolox/kg extract.
