INSTITUT TEKNOLOGI BANDUNG https://digilib.itb.ac.id/gdl/view/68610 Agarwood is a aromatic resin produced by Thymelaeaceae family that induced by stress. The formation of agarwood involve a complex process that rarely happend in nature. Futhermore, agarwood is harvested from plant’s branch, hence destructive to the plant, making the process takes a great amount of time. In addition, the population of the agarwood plant has been decreasing to the point that on 2019, Aquilaria sp and Gyrinops sp (agarwood main producer) has been listed in Appendix II by CITES. Biological elicitation using crude mycelial extract (CME) on agarwood callus is able to reduce the production time of agarwood. The aim of this research is to conduct an experiment on G. versteegii callus growth to arrange a growth curve, ergo, the stationary phase will be identified as the optimum elicitation time. In addition, a variation of CME concentration is used to determine the optimum consentration followed by it’s agarwood profile. A. niger is cultivied for 3 weeks in PDB. Callus culture is cultivied in Murashige-Skoog (MS) with half components media supplemented with NAA and BAP with 3:0,5 ppm w/w ratio, at room temperature and 100 rpm agitation. The growth curve showed that the log phase occur at day-0 up to day-10, the stationary phase at day- 10 to day 18, and lastly the death phase at day-18 to day 22. Extraction of agarwood by maseration on 100 rpm agitation using hexane for 3 days showed that the optimum CME consentration by order is 8 mg/L, 16 mg/L, and 4 mg/L with their respective yield 4,281%; 3,444%; and 2,608%. GC-MS analysist on agarwood sample that elicited with the optimum concentration (8 mg/L) detected 13 compounds from benze derivative, ester, and alcohol group with 1,2- Benzenedicarboxylic acid, bis(2-methylpropyl) ester (DIBP) as the key chemical compound with 17,584% of total agarwood. text
