Summary: | Gambier and tea are plants that have been used for generations as traditional medicine. The
development of technology and knowledge has led to an increase in research related to the
pharmacological activities of gambier and tea. One of them is the activity of plant extracts as
antimicrobials. In Indonesia, studies related to antimicrobial activity by isolates from these two
plants have not been widely carried out. This study was conducted to test the ability of catechin
from gambier and epicatechin and epigallocatechin gallate (EGCG) from tea as antimicrobial
compounds. The test was carried out using the microdilution method to determine the minimum
inhibitory concentration (MIC) and minimum bactericidal/fungicidal concentration (MBC/MFC)
against the bacteria Streptococcus mutans, Staphylococcus aureus, Escherichia coli, Pseudomonas
aeruginosa, Bacillus subtilis, and the fungi Aspergillus niger and Candida albicans. The MIC values
of the three compounds against bacteria and fungi were in the range of 500 to 1.000 µg/mL. The
MBC values for S. mutans were 4.000, > 4.000, and 4.000 µg/mL, respectively, for S. aureus were
1.000, 1.000, and 4.000 µg/mL, for E. coli and B. subtilis of the three compounds were 2.000 µg/mL,
and for P. aeruginosa at 4.000 µg/mL. The MFC value for A. niger of the three compounds was 1.000
µg/mL and for C. albicans was 500 µg/mL. It can be said that the isolates of catechins, epicatechins,
and EGCG have the potential as antimicrobial substances with moderate to mild strength. The MBC
or MFC/MIC ratio obtained showed that catechins, epicatechins, and EGCG had bactericidal
properties against S. aureus (except EGCG), E. coli, and B. subtilis as well as fungicidal properties
against A. niger and C. albicans. The three isolates against S. mutans and P. aeruginosa and EGCG
compounds against S. aureus can be said to be bacteriostatic.
