Summary: | Cavendish banana is a prime horticulture commodity, therefore its productivity
should be improved to meet the Indonesian community's needs and become a
foreign exchange. Conventionally, banana seedlings are produced uses few
saplings which are also prone to disease. One of the tools that can be used to
increase the production of healthy saplings is by using a tissue culture (in vitro)
technology, however this technology is costly. One of the solutions to reduce the
cost of production in the tissue culture method is by using an alternative medium
or combining the laboratory grade medium with a foliar fertilizer and organic
compounds to supply the insufficiency of nutrients. This study was conducted to
determine the best or optimum composition for the use of commercial leaf
fertilizer/PDK and young coconut water (coconut water) as an alternative for
Cavendish banana propagation based on several growth parameters used, namely
shoots number, length of pseudo-stem, leaves number and area, leaves "vigor",
roots number, and survival rate of Cavendish banana plantlets. This study used 9
treatment combinations namely P1 (MS0 +50 ml/L coconut water), P2 (MS0
+100 ml/L coconut water), P3 (1
2 MS0 + 1
2 PDK 50 ml/L coconut water), P4 (1
2
MS0 + 1
2 PDK +100 ml/L coconut water), P5 (1
4 MS0 + 3
4 PDK +50 ml/L coconut
water), P6 (1
4 MS0 + 3
4 PDK +100 ml/L coconut water), P7 (PDK+50 ml/L coconut
water), P8 (Full PDK +100 ml/L coconut water) which all of them were
supplemented with 0.5ml/L of shoot stimulant fertilizer/PPT, as well as a control
containing MS0. The result obtained in this study showed that the highest number
of shoots was obtained on a P4 medium. The longest pseudo-stem was obtained in
P3, P5, and MS0 medium. The highest number of leaves were obtained in P5
medium. The leaf "vigor" parameter was good for all plantlets, except for plantlets
grown in P7 and P8 medium. The biggest size of leaves was obtained in the P3
and P5 medium, and the highest number of roots was in P5 medium. All
Cavendish banana plantlets on optimization medium without and with addition of
PPT and coconut water survived after acclimatization, except in the preliminary
test which were grown on the highest concentration of PDK only. Therefore, it
can be concluded that the medium with a composition of 1
2 MS0 and 1
2 PDK and
100ml/L coconut water or 1
4 MS0 and 3
4 PDK and 50ml/L coconut water, can be
used as an alternative medium for Cavendish banana in vitro propagation |
