Microflora colonization is one of the causes of acne. This study aimed to determine the potential of ethanol extract and handeuleum (Graptophylum pictum (L.) Griff) leaf fraction in inhibiting acne-causing bacteria, Cutibacterium acnes and Staphylococcus aureus, and to isolate the active compounds. Extraction was carried out in two ways, maceration using 70% ethanol and infusion using distilled water. The selected extracts were fractionated using a liquid-liquid extraction method with nhexane, ethyl acetate, and ethanol-water as solvents. The selected fractions were further fractionated by classical column chromatography using the isocratic elution method. Purification was carried out by preparative thin layer chromatography method. The selected isolate candidates were tested for purity by single expansion thin layer chromatography method with three mobile phases. Antibacterial activity test was carried out qualitatively using agar disk diffusion methods and TLC-bioautography agar diffusion, and quantitatively by determining the minimum inhibitory concentration (MIC) using the microdilution method. MIC values for Cutibacterium acnes ATCC 11827 from ethanol extract, n-hexane fraction, ethyl acetate fraction, ethanol-water fraction, and isolate Z were 50; <0.78; 3.12; 100; >1 (mg/mL). While the MIC values for Staphylococcus aureus ATCC 6538 from ethanol extract, n-hexane fraction, ethyl acetate fraction, ethanol-water fraction, and isolate Z were 50; 6.25; 50; >100; 1 (mg/mL). The n-hexane fraction had better activity than ethanol extract and other fractions in inhibiting the growth of the two test bacteria. Isolate Z was isolated from the n-hexane fraction which has the potential as an antibacterial which is a steroid group compound. text
